Summary: | In this study, stainless steel plate (area of 5 cm2) was penetrated and platinized using electrophoretic deposition (EPD) procedure to create a porous, resistant and cohesive surface which subsequently used as disk for solid phase dynamic extraction (SPDE) method. The SPE disk was coated by polythiophene/multiwalled carbon nanotubes (PTh/MWCNTs) nanocomposite using in-situ electropolymerization method. The nanocomposite was characterized using scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FT-IR). A high surface area, porous and unbreakable platinized SPE disk was used for dynamic selective extraction, preconcentration and purification of thymol and carvacrol in thyme by high-performance liquid chromatography (HPLC) with UV detection. Influence of various variables including type of elution solvent, pH, ionic strength and extraction time was studied and optimized by central composite design (CCD). Under response surface methodology present method has admissible calibration curves over wide linear ranges of 0.01-100 µg mL-1 with high coefficient of determination (0.9998) that strongly confirm high applicability of present method for quantification of analyte, while their limit of detection (LODs) were 0.007 and 0.005 µg mL-1 and limit of quantification (LOQs) were 0.09 and 0.01 µg mL-1 for thymol and carvacrol, respectively. Also, enrichment factor (EF) was 87 for thymol and 54 for carvacrol and preconcentration factor was 142.8 for thymol and carvacrol. The SPDE-PTh/MWCNTs-disk-HPLC-UV was used for extraction and preconcentration of thymol and carvacrol in thyme samples and it satisfactory results were obtained. |
