Natural medicine such as keladi tikus, kemangi, and pepaya can be used as alternative medicine against cancer. The aim of this study was to evaluate the cytotoxic activity of keladi tikus, kemangi, and pepaya etanol extract against MCF-7 cells and determine of qualitative phytochemical constituent from the extracts. Samples powder are macerated with etanol 96 % for 72 hours. Samples were filtered, then filtrates are evaporated with rotary evaporator and put on waterbath to obtain crude extracts. Identification of pythochemical constituent with TLC using mobile phase n-heksan-aceton (7,5;2,5). Visualization is using annisaldeid-H2SO4, sitroborat, dragendorf, and FeCl3 spray reagent, then viewed under UV366 and daylight. Extract were diluted to obtain serial concentration of 500 ; 250 ; 125 ; 62,5 ; 31,25 µg/mL and tested the cytotoxic activity using MTT assay. Extract of keladi tikus, kemangi, and pepaya are positive contain alkaloid, phenolic, terpenoid, saponin, and flavonoid. The result of cytotoxic test show that extract of keladi tikus, kemangi, and pepaya have cytotoxic effect against MCF-7 cells with IC50 respectively, 17,40 ; 6,95 ; 9,73 µg/mL. it means that the leaf extracts are a potential source to be a anticancer agent.
