Summary:Yacon leaves (Smallanthus sonchifolius (Poepp. & Endl.) H. Robinson) contained caffeic, chlorogenic and ferulic acid. Previous study showed that these compounds may decrease blood glucose level. Formulating as tablet dosage could improve efficiency of yacon leaves application. This study was aimed to obtain an optimized formula of tablet using PVP as binder and natrium alginat as disintegrant. The study design was made by 5 formulas with various concentrations of PVP and sodium alginate (1%:7%; 2%:6%; 3%:5%; 4%:4%; 5%:3%). Parameter test was conducted on the flow rate, the angle of repose, tapped density, uniformity of weight, hardness, friability, and disintegration time. Optimization was designed by Design Expert 10.0.3.1 (trial) with simplex lattice design method which provides the optimum formula and predictive test results of physical properties of granules and tablets. Verification of the optimum formula then performed. The results of prediction and verification were analyzed using one sample t-test at 95% confidence level. The result showed that optimum formula of the tablet has obtained at a combination of 1% PVP and 7% sodium alginate. Statistical test on the prediction and verification data showed no significant differences in the angle of repose, tapped density, uniformity of weight, hardness, and disintegration time, so the data were valid. Significant differences showed in the flow rate and friability. However, the verification data on these parameters was eligible in the literature, so that the optimization results were valid.
