Summary: | Structure and development of sugarcane callus resulted by subculture on agar solidified Murashige & Skoog (MS) medium supplemented with 3 mg/1 2,4- Diclorophenoxyacetk acid (2,4-D) and 10 %of coconut milk has been observed. The callus inducted from young leaf explant of sugarcane var. Ps-851, incubated both the dark and underlight To observe the anatomical structure, callus obtained from 0-31 days incubation were fixed by 50 % FAA and cut using paraffin method Callus was stained with Mayer's Hemalum. Morphological observation showed that under light cultured callus has white-brownish color with mucilaginous texture. In other hand, the dark cultured callus has white-yellownish color with friable texture.S Anatomically, the calli had more meristematic cells in outside area than inside area. After subcultured, compact structure were formed on the outside and insides area. In darkness, compact structure on the outsides area developed into globular embryo like structure. Some calli with compact structure incubated on underlight, developed into meristemoid form, then meristematic dome and develop into shoot buds like structure at last. Morphologically, this structure had form green nodules in surface ofcalli. |
